Recombinant DNA Technology (BIO 680/560)

NOTE: The outline below contains two "discontinuities," so that the course consists of three blocks of experiments. The first and largest of these is devoted to the cloning, propagation and characterization of the lux operon, whereas the second block deals with the subcloning of the luxAB fragment encoding the luciferase enzyme. The final block of experiments concerns PCR amplification of luxA from natural isolates of bioluminescent bacteria and computer-aided analyses of their sequences.
