James Dunlop was born in Scotland.
James assumed the Lairdship of Dunlop after it was resigned by his uncle John (of Garnkirk) Dunlop. He too suffered persecution as a Presbyterian, being imprisoned for his beliefs from 1645 to 1667. In 1667, he won his freedom, but was forced to relinquish a portion of his lands to the Earl of Dundonald.
He married Elizabeth Cunningham.
